Ariad Pharmaceuticals, Inc. (NASDAQ:ARIA) today announced that Cancer Cell, one of the leading scientific journals, has published a comprehensive paper describing the design and preclinical characterization of an investigational kinase inhibitor (AP24534). The paper mentions AP24534 which is a first in class Pan BCR-ABL inhibitor used for the treatment of chronic myeloid leukemia, and the publication has been co-authored by Ariad scientists and other collaborating investigators from Oregon Health and Science University, Knight Cancer Institute and the Howard Hughes Medical Institute. The data demonstrates inhibition of all known variants of the CML target protein.

Ariad Pharmaceuticals has been working very hard to transform the lives of patients suffering from cancer by coming up with breakthrough medicines. Ariad scientists first determined the structure of the T3151 mutant of BCR-ABL to discover how the mutation changes the drug binding site.

The publication for the very first time clearly described the chemical structure of AP24534 and its activity against all BCR-ABL mutants, including the T3151 mutant that does not react to the currently available therapies for chronic myeloid leukemia (CML). AP24534 was discovered by the Ariad Pharmaceuticals scientists and currently it is in Phase 1 dose-escalating clinical trial in patients with refractory CML, acute myeloid leukemia and other hematological malignancies. As per the publication, treatment with BCR-ABL inhibitors is effective initially with patients suffering from chronic myeloid leukemia but it can result in the emergence of BCR-ABL mutations that confer drug resistance over a period of time.
